Study on the improvement and mechanism of curcumin on lipid deposition in HepG2 cells
Objective:,2,To investigate the effects of curcumin on oleic acid induced lipid deposition in HepG2 cells, as well as its mechanism.,Methods:,2,HepG2 cells were divided into the control group, model group, and curcumin low-and high-dose groups.Except the control group, the other groups were treated with 0.3 mmol/L oleic acid for 24 hours to induce cell lipid deposition model.After the model was established, curcumin intervention groups were treated with 50 and 100 μmol/L curcumin for 9 hours, respectively.Lipid deposition was observed by oil red O staining.The contents of triglyceride(TG) and total cholesterol(TC) were detected by the kits.The mRNA expressions of sterol-regulatory element binding protein 1C(,SREBP1C,), fatty acid synthetase(,FAS,) and acetyl CoA carboxylase α(,ACCα,) were detected by PCR.,Results:,2,Compared with the control group, the lipid droplets in the model group were significantly increased, the levels of TG and TC were significantly increased(,P,<,0.05,P,<,0.01), and the mRNA expression levels of SREBP1C and its downstream genes ,FAS, and ,ACCα, were significantly increased(,P,<,0.01) .Compared with the model group, the red lipid droplets in the curcumin intervention groups were significantly reduced, the TG level was significantly decreased(,P,<,0.01), and the mRNA expression levels of ,SREBP1C,FAS, and ,ACCα, were significantly decreased(,P,<,0.05,P,<,0.01) .The improvement on lipid deposition was more significantly in the curcumin low-dose(50 μmol/L) group.,Conclusion:,2,Curcumin may inhibit the lipid synthesis by down-regulating the expressions of SREBP1C, FAS and ACCα, and thus improve the lipid deposition of HepG2 cells.
