目的:,2,探讨生地黄多糖对环磷酰胺诱导的免疫抑制模型小鼠的免疫调节作用。,方法:,2,将60只雄性Balb/c小鼠随机分为对照组、模型组、阳性对照组以及生地黄多糖低、中、高剂量组,每组10只。除对照组外,其他各组小鼠腹腔注射80 mg/kg环磷酰胺,每日1次,连续2 d。实验第3天起,阳性对照组小鼠灌胃给予50 mg/kg香菇多糖溶液,生地黄多糖各剂量组小鼠分别灌胃给予5、10、20 g/kg生地黄多糖溶液,连续7 d。末次给药后,取血,分离肝脏、脾脏、胸腺;碳粒廓清实验评价单核细胞吞噬能力;血细胞分析仪检测白细胞数、红细胞数、血小板数、淋巴细胞数、中性粒细胞数、单核细胞数;ELISA检测血清肿瘤坏死因子-α(TNF-α)、γ-干扰素(IFN-γ)、白细胞介素(IL)-4、IL-10、IL-17水平;流式细胞术检测脾脏淋巴细胞CD3,+,CD4,+,与CD3,+,CD8,+,细胞的比例。,结果:,2,与对照组相比,模型小鼠的体质量、脾脏指数和胸腺指数明显下降(,P,<,0.05,,P,<,0.01),廓清系数K和吞噬指数α显著降低(,P,<,0.05);体内白细胞数、红细胞数、血小板数、淋巴细胞数、中性粒细胞数均明显减少(,P,<,0.01),而单核细胞数显著升高(,P,<,0.05);血清TNF-α、IL-17水平显著升高(,P,<,0.01),而IFN-γ和IL-4水平显著降低(,P,<,0.05,,P,<,0.01);CD3,+,CD4,+,细胞表达明显降低(,P,<,0.01),CD3,+,CD8,+,细胞表达明显升高(,P,<,0.05),CD3,+,CD4,+,/CD3,+,CD8,+,比值显著降低(,P,<,0.01)。与模型组相比,生地黄多糖各剂量组小鼠的脾脏指数明显升高(,P,<,0.01),高剂量组的廓清系数K和吞噬指数α均明显升高(,P,<,0.05,,P,<,0.01),白细胞数、血小板数、淋巴细胞数、中性粒细胞数均显著升高(,P,<,0.05,,P,<,0.01)。生地黄多糖作用可显著降低模型小鼠血清TNF-α、IL-17水平,促进IFN-γ、IL-4、IL-10表达(,P,<,0.05,,P,<,0.01);同时上调脾淋巴细胞CD3,+,CD4,+,细胞的表达水平,降低CD3,+,CD8,+,细胞表达水平,提高CD3,+,CD4,+,/CD3,+,CD8,+,比值(,P,<,0.05,,P,<,0.01)。,结论:,2,生地黄多糖能够增强环磷酰胺诱导的免疫抑制小鼠的免疫功能。
Objective:,2,To study the immunomodulatory effects of Radix Rehmanniae Recens polysaccharide on cyclophosphamide-induced immunosuppression in mice.,Methods:,2,Sixty male Balb/c mice were randomly divided into the control group, model group, positive control group and Radix Rehmanniae Recens polysaccharide groups with low-, middle-and high-dose, 10 mice in each group.Except the control group, mice in the other groups were intraperitoneally injected with cyclophosphamide(80 mg/kg) once a day for 2 days.From the third day of experiment, mice in the positive control group were treated with 50 mg/kg lentinan by intragastric administration, and mice in the Radix Rehmanniae Recens polysaccharide groups with different doses were treated with 5, 10 and 20 g/kg Radix Rehmanniae Recens polysaccharide solution for 7 days, respectively.After the last administration, the blood was taken, the liver, spleen and thymus were separated.The phagocytosis of monocytes was evaluated by carbon clearance test.The counts of white blood cells, red blood cells, platelets, lymphocytes, neutrophils and monocytes were detected by hematology analyzer.The serum levels of tumor necrosis factor-α(TNF-α), interferon-γ(IFN-γ), interleukin(IL) -4, IL-10 and IL-17 were detected by ELISA.The proportions of CD3,+,CD4,+, and CD3,+,CD8,+, cells in spleen lymphocytes were detected by flow cytometry.,Results:,2,Compared with the control group, the body mass, spleen index and thymus index of the model mice were significantly decreased(,P,<,0.05,P,<,0.01), the clearance coefficient K and phagocytosis index α were significantly decreased(,P,<,0.05) ; the counts of white blood cells, red blood cells, platelets, lymphocytes and neutrophils were significantly decreased(,P,<,0.01), while the count of monocytes was significantly increased(,P,<,0.05) ; the serum levels of TNF-α and IL-17 were significantly increased(,P,<,0.01), while the levels of IFN-γ and IL-4 were significantly decreased(,P,<,0.05,P,<,0.01) ; the expression of CD3,+,CD4,+, cells was significantly decreased(,P,<,0.01), the expression of CD3,+,CD8,+, cells was significantly increased(,P,<,0.05), and the ratio of CD3,+,CD4,+,/CD3,+,CD8,+, was significantly decreased(,P,<,0.01) .Compared with the model group, the spleen index of mice in the Radix Rehmanniae Recens polysaccharide groups with different doses was significantly increased(,P,<,0.01), the clearance coefficient K and phagocytosis index α of mice in the high-dose group were significantly increased(,P,<,0.05,P,<,0.01), and the counts of white blood cells, platelets, lymphocytes and neutrophils were significantly increased(,P,<,0.05,P,<,0.01) .Radix Rehmanniae Recens polysaccharide could significantly reduce the serum levels of TNF-α and IL-17 and promote the expressions of IFN-γ, IL-4 and IL-10 in model mice(,P,<,0.05,P,<,0.01) ; meanwhile, the expression level of CD3,+,CD4,+, cells was up-regulated, the expression level of CD3,+,CD8,+, cells was decreased, and the ratio of CD3,+,CD4,+,/CD3,+,CD8,+, was increased(,P,<,0.05,P,<,0.01) .,Conclusion:,2,Radix Rehmanniae Recens polysaccharide can enhance the immune function of the immunosupression mice induced by cyclophosphamide.
