目的,2,研究院内制剂泌尿排石合剂对大鼠草酸钙肾结石形成的影响,并初步探讨其机制。,方法,2,将SPF级健康雄性SD大鼠40只随机分为正常组、模型组、阳性对照组(枸橼酸钾)、泌尿排石合剂组,每组10只。采用1%乙二醇自由饮水和2%氯化铵灌胃建立大鼠草酸钙肾结石模型,各组给予相应药物干预。观察各组大鼠24 h尿量、尿草酸含量、尿Ca,2+,、P,3+,、Mg,2+,水平及血清BUN、Cr、Ca,2+,、P,3+,、Mg,2+,水平;显微镜下观察肾组织切片中草酸钙结晶沉积及病理变化。,结果,2,①24 h尿量、血生化指标:泌尿排石合剂组和阳性对照组24 h尿量高于正常组和模型组(,P,<,0.05);与模型组比较,泌尿排石合剂组和阳性对照组24 h尿草酸、血尿Ca,2+,及P,3+,水平明显降低(,P,<,0.05);且泌尿排石合剂组较阳性对照组更低(,P,<,0.05)。②血尿Mg,2+,水平比较:阳性对照组与泌尿排石合剂组高于模型组(,P,<,0.05);阳性对照组与泌尿排石合剂组血尿Mg,2+,水平比较,差异无统计学意义(,P,>,0.05)。③肾功能:泌尿排石合剂组与阳性对照组较模型组明显改善(,P,<,0.05)。④病理变化:泌尿排石合剂组与阳性对照组较模型组草酸钙结晶在肾组织中沉积面积明显减少,肾小管水肿减轻。,结论,2,泌尿排石合剂能促进草酸排泄、降低血尿钙磷水平、增加血镁水平,从而抑制实验性大鼠体内肾草酸钙晶体的形成,最终发挥保护肾脏功能的作用。
Objective: To investigate the effect of Miniao Paishi Mixture of our hospital on the formation of calcium oxalate renal calculi in rats, and explore its mechanism preliminarily.,Methods,2,Forty healthy male SD rats of SPF grade were randomly divided into normal group, model group, positive control group (potassium citrate) and Miniao Paishi Mixture group, 10 in each group. To establish calcium oxalate kidney stone model in rats, 1% ethylene glycol was given with drinking water, and 2% ammonium chloride was given by gavage. And each group was given corresponding drugs intervention. The 24 h urine volume, urinary oxalic acid content, urinary Ca,2+,, P,3+,, Mg,2+, levels and serum BUN, Cr, Ca,2+,, P,3+,, Mg,2+, levels of rats in each group were observed. Calcium oxalate crystal deposition and pathological changes in renal tissue sections were observed under microscope.,Results,2,①The 24 h urine volume and blood biochemical indexes: the 24 h urine volume of Miniao Paishi Mixture group and positive control group was higher than that of normal group and model group (,P,<,0.05). Compared with the model group, the levels of 24 h urinary oxalic acid, Ca,2+, and P,3+, in the blood and urine in the Miniao Paishi Mixture group and the positive control group were significantly lower (,P,<,0.05). Moreover, those in the Miniao Paishi Mixture group was lower than the positive control group (,P,<,0.05). ②Comparison of serum and urinary Mg,2+, levels: the positive control group and Miniao Paishi Mixture group were higher than model group (,P,<,0.05). There was no significant difference in Mg,2+, level in blood and urine between positive control group and Miniao Paishi Mixture group (,P,>,0.05). ③Renal function: the Miniao Paishi Mixture group and positive control group were significantly better than the model group (,P,<,0.05). ④Pathological change: compared with the model group, the deposition area of calcium oxalate crystals in the renal tissue was significantly reduced and the renal tubular edema decreased in the Miniao Paishi Mixture group and the positive control group.,Conclusion,2,Miniao Paishi Mixture can promote the excretion of oxalic acid, decrease the levels of calcium and phosphorus in blood and urine, increase the level of magnesium in blood, thus inhibit the formation of renal calcium oxalate crystal in experimental rats, and finally play a role in protecting renal function.
