目的,2,观察四氯化碳(CCl,4,)及二甲基亚硝胺(DMN)诱导小鼠肝纤维化模型肝脏淋巴细胞亚群变化特点,并进一步观察中药扶正化瘀方对淋巴细胞亚群变化的调节作用。,方法,2,分别采用10%CCl,4,及0.5%DMN腹腔注射制备小鼠肝纤维化模型,观察小鼠肝脏大体观及脏器指数,检测小鼠血清ALT、AST表达变化,HE和天狼猩红染色观察肝组织炎症及纤维化程度,流式细胞术观察小鼠肝脏淋巴细胞亚群比例和数量变化。,结果,2,与正常组比较,两种模型组小鼠肝脏表面粗糙,质地变硬,粟粒样表现明显,肝体比下降,脾体比升高,血清ALT、AST表达增加,肝组织病理染色显示明显的炎症和胶原沉积,肝脏NK细胞、NKT细胞、CD4,+,T细胞比例和数量下降,CD8,+,T细胞和B细胞比例和数量升高(,P,<,0.05,,P,<,0.01);与模型组比较,药物组小鼠肝脏大体观明显改善,肝体比增加,脾体比下降,血清ALT、AST表达下降,肝组织炎症和胶原沉积明显减轻,肝脏NK细胞、NKT细胞、CD4,+,T细胞比例和数量升高,CD8,+,T细胞和B细胞比例和数量下降(,P,<,0.05,,P,<,0.01)。,结论,2,CCl,4,和DMN诱导肝纤维化模型小鼠肝脏淋巴细胞亚群免疫紊乱,中药扶正化瘀方可调节肝脏免疫微环境,纠正免疫紊态,发挥抗肝纤维化作用。
