目的,2,基于雌二醇的变化探讨淫羊藿总黄酮胶囊(EFC)对去卵巢骨质疏松模型大鼠的作用机制。,方法,2,将48只3月龄SD大鼠随机分为4组,即假手术组(SHAM组)、模型组(OVX组)、雌二醇组(E,2,组)、EFC组,每组12只。通过行双侧卵巢切除术造模,造模成功后E,2,组及EFC组每天分别给予相应药物干预,SHAM组以及OVX组每天给予等量的0.9%氯化钠溶液,连续12周。通过双能X线吸收测量法观察EFC对大鼠腰椎和股骨骨密度的影响,Micro-CT检测大鼠骨小梁微结构的变化,ELISA法检测大鼠血清中E,2,、Ⅰ型前胶原N端前肽(PINP)、Ⅰ型胶原羧基末端肽(β-CTX)含量变化。,结果,2,①与OVX组比较,EFC组血清E,2,含量显著增高(,P,<,0.01),但含量低于E,2,组;②与OVX组比较,EFC组血清β-CTX含量明显降低(,P,<,0.01),血清PINP含量上升(,P,<,0.05);③EFC组腰椎骨密度、股骨骨密度较OVX组有所提高,但无统计学意义;④与OVX组比较,EFC组骨小梁体积分数增高(,P,<,0.05),骨小梁数明显增加(,P,<,0.01),骨小梁厚度有所提高,但无统计学意义,骨小梁分离度明显降低(,P,<,0.01);⑤E,2,与PINP无显著相关性(,P,>,0.05),E,2,与β-CTX呈显著性负相关(,r,=-0.6596,,P,<,0.001)。,结论,2,EFC可能发挥雌激素样作用,通过调节血清骨代谢标志物含量以及骨小梁微结构的变化,改善骨密度从而起到防治骨质疏松的作用。
Objective: To explore the mechanism of Epimedium Total Flavonoid Capsule(EFC)on ovariectomized osteoporosis model rats.,Methods,2,Forty-eight 3-month-old SD rats were randomly divided into 4 groups: SHAM operation group (Sham group), model group(OVX group), estradiol group(E,2, group)and EFC group, 12 rats in each group. The model was established by bilateral oophorectomy. After successful modeling, E,2, group and EFC group were given corresponding drug intervention respectively every day, while SHAM group and OVX group were given the same amount of 0.9% NaCl solution every day, each group was administered for 12 consecutive weeks. The effects of EFC on bone mineral density of lumbar vertebrae and femur in rats were observed by dual energy X-ray absorption measurement, the changes of trabecular microstructure in rats were detected by Micro-CT, and the content changes of E,2,, type Ⅰ procollagen amino terminal peptide(PINP) and type Ⅰ collagen carboxyl terminal peptide(β-CTX) in rats’ serum were detected by ELISA.,Results,2,①Compared with OVX group, serum E,2, content in EFC group was significantly increased (,P,<,0.01), but lower than that in E,2, group. ②Compared with OVX group, serum β-CTX content in EFC group was significantly decreased (,P,<,0.01), and serum PINP content was increased (,P,<,0.05). ③The bone mineral density of lumbar vertebrae and femur in EFC group were higher than those in OVX group, but there was no statistical significance. ④Compared with OVX group, the volume fraction of bone trabecula in EFC group was increased (,P,<,0.05), the number of bone trabecular was increased significantly (,P,<,0.01), the thickness of bone trabecular was increased, but there was no statistical significance, and the separation degree of bone trabecular was decreased significantly (,P,<,0.01). ⑤There was no significant correlation between E,2, and PINP (,P,>,0.05), but there was a significant negative correlation between E,2, and β-CTX(,r,=-0.659 6, ,P,<,0.001).,Conclusion,2,EFC may play an estrogen-like role in the prevention and treatment of osteoporosis by regulating the content of serum bone metabolism markers and the changes of bone trabecular microstructure and improving bone density.
