目的,2,探究天麻醇提物降低短暂性大脑中动脉阻塞(tMCAO)模型大鼠出血转化(HT)的作用。,方法,2,SD大鼠随机分为正常组、模型tMCAO 3 h-HT组、模型tMCAO 6 h-HT组、天麻tMCAO 3 h-HT组及天麻tMCAO 6 h-HT组,天麻组大鼠于术前连续7 d灌胃天麻醇提物(843.75 mg/kg,10 ml/kg),其余各组给予相同体积溶媒。采用线栓法建立大鼠tMCAO模型,分别于缺血后3 h、6 h拔出线栓建立tMCAO 3 h-HT、tMCAO 6 h-HT模型。术后48 h检测脑组织HT发生率、出血量评分、血红蛋白含量、死亡率及脑组织微管相关蛋白-2(MAP-2)的表达。,结果,2,①与模型组比较,天麻组大鼠的死亡率及HT的发生率有所下降,但差异无统计学意义(,P,>,0.05);②与模型组比较,天麻醇提物能显著降低tMCAO 3 h-HT、tMCAO 6 h-HT模型大鼠脑组织的出血量评分(,P,<,0.05,,P,<,0.01),并降低tMCAO 3 h-HT模型大鼠的血红蛋白含量(,P,<,0.05);③与模型组比较,天麻醇提物可缩小tMCAO 6 h-HT模型大鼠缺血中心区的面积(,P,<,0.05),并增大缺血半暗带区的面积(,P,<,0.05)。,结论,2,天麻醇提物预防性给药7 d后可减少tMCAO大鼠HT,还可能具有延长溶栓时间窗的潜在作用。
Objective: To explore the effect of Gastrodiae Rhizoma alcohol extract on reducing hemorrhagic transformation (HT) in transient middle cerebral artery occlusion (tMCAO) model rats.,Methods,2,SD rats were randomly divided into normal group, model group, model tMCAO 3 h-HT group, model tMCAO 3 h-HT group, Gastrodiae Rhizoma tMCAO 3 h-HT group, and Gastrodiae Rhizoma tMCAO 6 h-HT group. The rats in Gastrodiae Rhizoma group were given Gastrodiae Rhizoma alcohol extract (843.75 mg/kg, 1 ml/100 g) by gavage for 7 d before operation. The rats in other groups were given the same volume of solvent. The tMCAO models of rats were established by suture method. The tMCAO 3 h-HT and tMCAO 6 h-HT models were established by pulling out thread at 3 h and 6 h after ischemia respectively. HT incidence, bleeding volume score, hemoglobin content, mortality and expression of microtubule associated protein-2 (MAP-2) in brain tissue were measured 48 h after operation.,Results,2,①Compared with the model group, the mortality rate and HT incidence of rats in the Gastrodiae Rhizoma groups were reduced, but the difference was not statistically significant (,P,>,0.05). ②Compared with the model group, the alcohol extract of Gastrodiae Rhizoma significantly reduced the bleeding volume score (,P,<,0.05, ,P,<,0.01) in the brain tissue of tMCAO 3 h-HT and tMCAO 6 h-HT model rats, and reduced the hemoglobin content (,P,<,0.05) of tMCAO 3 h-HT model rats. ③Compared with the model group, Gastrodiae Rhizoma alcohol extract reduced the area of the ischemic central zone (,P,<,0.05) and increased the area of the ischemic semidark zone (,P,<,0.05) in the tMCAO 6 h-HT model rats.,Conclusion,2,Prophylactic administration of Gastrodiae Rhizoma ethanol extract for 7 d can reduce HT in tMCAO model rats, and may has the potential to prolong the thrombolytic time window.
