目的,2,考察原儿茶醛脑微透析探针的体内外回收率及其稳定性。,方法,2,通过高效液相色谱法检测微透析样品中原儿茶醛的浓度。考察不同灌流速度、药物浓度对原儿茶醛脑微透析探针体内外正向和反向回收率的影响,以及体内外回收率的稳定性。,结果,2,原儿茶醛脑探针体外回收率基本相同,并且体外和体内透析法的相对回收率亦基本相同,因此可采用体内反向回收率作为药物的探针回收率来校正。探针回收率与流速(1.0、1.5、2.0、2.5、3.0 μl/min)成反比,与原儿茶醛药物浓度(3.13、6.25、12.50、25.00 μg/ml)无关。原儿茶醛体内外脑探针8 h内回收率的变化基本保持稳定。,结论,2,建立的脑微透析方法可用于原儿茶醛大鼠脑药动学研究。
Objective: ,To investigate the ,in vitro ,and, in vivo, recovery rates and stability of protocatechuic aldehyde brain microdialysis probe.,Methods,2,The concentration of protocatechuic aldehyde in microdialysis samples was determined by high performance liquid chromatography. The effects of different drug perfusion speeds and concentrations on forward and reverse recovery of protocatechuic aldehyde microdialysis brain probe, in vitro ,and, in vivo,, and the stability of recovery ,in vitro ,and, in vivo, were investigated.,Results,2,The ,in vitro, recovery of the protocatechuic aldehyde brain probe was basically the same, and the relative recoveries of the ,in vitro ,and ,in vivo ,dialysis methods were also basically the same, so the ,in vivo ,reverse recovery can be used as the probe recovery of the drug for calibration. The recovery rate of the probe was inversely proportional to the flow rates (1.0, 1.5, 2.0, 2.5, and 3.0 μl/min) and has nothing to do with the concentration of protocatechuic aldehyde (3.13, 6.25, 12.50, 25.00 μg/ml). The ,in vivo ,and, in vitro, recovery rate of protocatechuic aldehyde brain probes within 8 h remained basically stable.,Conclusions,2,The established protocatechuic aldehyde brain microdialysis method can be used for brain pharmacokinetic study in rats.
