目的,2,建立同时测定余甘子总酚中没食子酸和葡糖倍苷的UPLC含量测定方法,并进行余甘子总酚的UPLC指纹图谱研究。,方法,2,采用CAPCELL PAK C,18, MG(4.6 mm × 150 mm,5 μm)色谱柱;流动相:甲醇-2%乙酸水溶液,梯度洗脱;检测波长:273 nm;柱温:30 ℃;流速:0.8 ml/min。,结果,2,没食子酸和葡糖倍苷分别在5.82~465.20 μg/ml、12.38~793.00 μg/ml范围内呈良好线性关系;平均加样回收率分别为100.57%和99.22%,,RSD,均,<,3.0%;指纹图谱共标示出7个特征峰,相似度,>,0.90。,结论,2,所建立的方法简便、结果准确、重复性好,可作为余甘子总酚原料的质量控制方法。
objective: ,To establish a UPLC assay method for the determination of gallic acid and glucose times nucleoside and a UPLC fingerprint analysis method for total phenol of Phyllanthi Fructus.,Methods,2,The analysis conditions were set at a CAPCELL PAK C,18, MG(4.6 mm × 150 mm,5 μm)chromatographic column; mobile phase: methanol-2% acetic acid aqueous solution, gradient elution; detection wavelength: 273 nm; column temperature: 30 ℃; flow rate: 0.8 ml/min.,Results,2,Good linear relationship was observed for gallic acid in the range of 5.82~465.20 μg/ml and glucose times nucleoside in the range of 12.38~793.00 μg/ml respectively. And the average recovery rates of gallic acid and glucoside were 100.57% and 99.22% respectively, with the ,RSD,s both less than 3.0%. The fingerprint showed 7 characteristic peaks, and the similarity of fingerprint was more than 0.90.,Conclusion,2,This method is simple, accurate and reproducible, and could be used as a quality control method for total phenol of Phyllanthi Fructus.
