摘要:The incidence of breast cancer is increasing year by year and now ranks first in women malignant tumors. In recent years, a lot of progress has been made in the research related to the treatment of breast cancer with traditional Chinese medicine (TCM). In addition, the individualized treatment principle of TCM surgery, which combines disease differentiation and syndrome differentiation, holistic and localized, internal and external treatment, is consistent with the precise treatment concept based on different molecular subtypes in modern medicine. In this paper, in view of the different molecular subtypes and treatment status of breast cancer, the important research achievements of TCM researches in recent years have been summarized from anti-triple-negative breast cancer (TNBC) recurrence and metastasis, improvement of human epidermal growth factor receptor 2 (HER2)-positive breast cancer targeted drug resistance, and alleviation of hormone receptor (HR)-positive breast cancer endocrine therapy resistance and its adverse reactions with TCM. It will provide the development direction for TCM treatment of breast cancer.

摘要:Objective: One kind of biomimetic nanoparticles was constructed to deliver quercetin (QT) in an attempt to improve the targeting efficiency, so as to improve the inhibitory and apoptosis effect of QT for colorectal cancer (CRC) cells.MethodsPLGA.QT was prepared by emulsification-solvent evaporation method, the surface of which was coated with red blood cell membrane (RBC), aminoethyl anisamide (AEAA) was then modified on PLGA.QT using “post-insertion” method, and biomimetic targeting nanoparticles (PLGA.QT.RBC-AEAA) were finally obtained. Then, the particle size, zeta potential, encapsulation rate, loading capacity and drug release of PLGA.QT.RBC-AEAA nanoparticles were characterized. The cellular uptake of the nanoparticles was observed by confocal laser microscopy, and their inhibition effect of proliferation and apoptosis on mouse CRC CT26 cells were evaluated by MTT assay and flow cytometry respectively.ResultsThe size, zeta potential, encapsulation rate, and loading capacity of PLGA.QT.RBC-AEAA nanoparticles were (107.3±7.7) nm, (-17.5±0.6) mV, (65.7±5.2)% and (4.8±0.2)%, respectively. The nanoparticles exhibited small, spherical and uniform particle size, which could effectively encapsulate QT. In vitro release experiments showed that PLGA.QT.RBC-AEAA nanoparticles had faster drug release rate in acidic environment than in neutral environment (P<0.01). In addition, the biomimetic targeting nanoparticles could promote more QT to enter into cells (P<0.05), and effectively enhance its toxicity on CT26 cells (P<0.01). Compared with the free drug, the inhibition rate of CT26 cell proliferation by biomimetic targeting nanoparticles was increased nearly 3 times. Meanwhile, compared with the non-targeting group, the targeting nanoparticles had a stronger pro-apoptotic effect (P<0.01).ConclusionThe PLGA.QT.RBC-AEAA nanoparticles which modified with AEAA targeting ligand can effectively improve the targeted delivery efficiency of QT, enhance its inhibitory and apoptosis effect on CRC cells, which can provide an experimental basis for the evaluation of QT in vivo pharmacokinetics and pharmacodynamics in the future.
摘要:Objective: To observe the clinical efficacy of chemotherapy combined with modified Yiyanxiao Formula in the treatment of pancreatic cancer with peritoneal metastasis.MethodsSixty-eight patients of pancreatic cancer with peritoneal metastasis were enrolled and randomly divided into the control group and treatment group, 34 cases in each group. The patients in both groups were treated with gemcitabine intravenous drip combined with cisplatin intraperitoneal perfusion chemotherapy. Twenty-one was a course of treatment and a total of two courses of treatment was performed. Based on above, the patients in the treatment group were treated with modified Yiyanxiao Formula orally. Before and after treatment, the levels of the tumor markers including carbohydrate antigen 199 (CA199), carcinoembryonic antigen (CEA) and carbohydrate antigen 125 (CA125), and the inflammatory cytokines including interleukin-1β(IL-1β), tumor necrosis factor-α (TNF-α) and IL-6 in the two groups were detected and compared. The clinical efficacy of the two groups was evaluated. The total scores of traditional Chinese medical symptoms and the incidence of adverse reactions were compared between the two groups.Results①After treatment, the disease control rates of the treatment group and the control group were 64.7% and 44.1% respectively, and the disease control rate of the treatment group was higher than that of the control group (P<0.05). The disease remission rates of the treatment group and the control group were 8.8% and 5.9% respectively, with no statistically significant difference between the two groups (P>0.05). ②After treatment, the total scores of traditional Chinese medical symptoms in both groups were decreased compared with those before treatment (P<0.05), and the score of the patient in the treatment group was lower than that in the control group (P<0.05). ③After treatment, the levels of CA199 and CEA in the two groups were significantly decreased compared with those before treatment (P<0.05), and the level of CA199 in the treatment group was lower than that in the control group (P<0.05). ④After treatment, the levels of IL-1β and IL-6 in the two groups were decreased compared with those before treatment (P<0.05), the level of TNF-α in the treatment group was also decreased compared with that before treatment (P<0.05), and the levels of IL-1β and IL-6 in the treatment group were lower than those in the control group (P<0.05). ⑤During the treatment, the incidence of adverse reactions was 44.1% in the treatment group and was 76.5% in the control group. The incidence of adverse reactions in the treatment group was significantly lower than that in the control group (P<0.05).ConclusionChemotherapy combined with modified Yiyanxiao Formula can effectively inhibit the expressions of tumor markers and inflammatory cytokines in the treatment of patients of pancreatic cancer with peritoneal metastasis, improve the clinical symptoms of the patients, increase the disease control rate, and decrease the incidence of adverse reactions of chemotherapy.

摘要:Objective: To explore the influence of Ginkgo diterpenoid lactone glucosamine injection on the level of serum galectin-3 (gal-3) in patients with acute ischemic stroke (AIS), and observe the relationship between the changes of serum gal-3 level and the neurological deficit symptoms and prognosis of the patients.MethodsA total of 88 patients with AIS were enrolled and randomly divided into the control group and trial group, 44 cases in each group. The patients in both groups were orally treated with aspirin enteric-coated tablets. Based on above, the patients in the trial group were treated with intravenous drip of Ginkgo diterpenoid lactone glucosamine injection, and the patients in the control group were treated with intravenous drip of placebo injection. The treatment course was 14 d. Before and after treatment, the levels of serum gal-3, interleukin-1β (IL-1β), IL-6 and tumor necrosis factor-α (TNF-α) in the two groups were detected and compared. Before treatment and on the 14th, 28th and 90th day of treatment, the scores of National Institutes of Health Stroke Scale (NIHSS) in the two groups were evaluated and compared. Before treatment and on the 90th day of treatment, the scores of modified Rankin Scale (mRS) in the two groups were evaluated and compared. The correlation between the changes of serum gal-3, IL-1β, IL-6 and TNF-α levels and the poor prognosis (mRS scores≥3 points on the 90th day of treatment) was analyzed.ResultsDuring the study, 8 patients in the control group were excluded or dropped out, and 8 patients in the trial group were excluded or dropped out. Finally, 36 cases in the control group and 36 cases in the trial group were included in the statistical analysis. ①On the 14th, 28th and 90thday of treatment, the scores of NIHSS in the two groups were significantly decreased compared with those before treatment (P<0.05), and the score of the trial group was lower than that of the control group (P<0.05). ②On the 90th day of treatment, the scores of mRS in the two groups were significantly decreased compared with those before treatment (P<0.05), and the score of the trial group was lower than that of the control group (P<0.05). ③After treatment, the serum gal-3 level of the trial group was significantly decreased compared with that before treatment (P<0.05), the serum gal-3 level of the control group was significantly increased compared with that before treatment (P<0.05), and the serum gal-3 level of the trial group was significantly lower than that of the control group (P<0.05). ④After treatment, the serum IL-1β level of the trial group was significantly decreased compared with that before treatment (P<0.05), and the TNF-α level was significantly increased (P<0.05). ⑤Poor prognosis was positively correlated with the increased serum gal-3 and IL-1β levels (P<0.05), and poor prognosis was negatively correlated with the increased serum TNF-α level (P<0.05).ConclusionGinkgo diterpenoid lactone glucosamine injection can effectively improve the symptoms and prognosis of neurological deficits in patients with AIS, and its mechanism may be related to the reduction of the serum gal-3 level in patients to achieve neuroprotection.
摘要:Objective: To observe the effect of electroacupuncture (EA) intervention on the mental symptoms and quality of life in amphetamine-type stimulant (ATS) abstainers.MethodsA total of 88 ATS abstainers were included and randomly divided into the conventional treatment group and EA treatment group, 44 cases in each group. After acute detoxification, the patients in the conventional treatment group were given natural withdrawal therapy on the basis of health education. The patients in the EA treatment group were given EA therapy based on the routine intervention, 3 times every week and a total of 10 times treatment were performed. Before treatment, after 1 and 2 weeks of treatment, and at the end of treatment, the scores of amphetamine withdrawal questionnaire (AWQ), positive and negative syndrome scale (PANSS) and quality of life for drug addicts scale (QOL-DA) were assessed and compared between the two groups.ResultsDuring the study, 1 case in the conventional treatment group was eliminated, finally 43 cases in the conventional treatment group and 44 cases in the EA treatment group were included in the statistical analysis. ①After 1 and 2 weeks of treatment and at the end of treatment, the AWQ score of the EA treatment group was decreased compared with that before treatment (P<0.05). After 2 weeks of treatment and at the end of treatment, the AWQ score of the EA treatment group was lower than that of the conventional treatment group (P<0.05). ②After 1 and 2 weeks of treatment and at the end of treatment, the total score of PANSS and the scores of positive scale and general psychopathological scale in the two groups were decreased compared with those before treatment (P<0.05), the score of negative scale in PANSS of the EA treatment group was also decreased compared with that before treatment (P<0.05), and the positive scale score of the EA treatment group was lower than that of the conventional treatment group (P<0.05). After 2 weeks of treatment and at the end of treatment, the score of negative scale in PANSS of the conventional treatment group was decreased compared with that before treatment (P<0.05), and the total score and the scores of negative scale and general psychopathological scale in the EA treatment group were lower than those in the conventional treatment group (P<0.05). ③After 1 and 2 weeks of treatment and at the end of treatment, the QOL-DA score of the EA treatment group was increased compared with that before treatment (P<0.05), and the score of the EA treatment group was higher than that of the conventional treatment group (P<0.05).ConclusionEA therapy can improve the mental symptoms and the quality of life for ATS abstainers with short onset time, and its curative effect gradually increases with the extension of treatment.

摘要:Objective: To establish the characteristic fingerprint and content determination methods of Senecionis Scandentis Herba Formula Granules (SSHFG), thus to provide reference for its quality evaluation.MethodsThe characteristic fingerprint of SSHFG was developed by using a CAPCELL PAK C18 AQ column (4.6 mm × 250 mm, 5.0 μm) with gradient elution of acetonitrile-0.2 % phosphoric acid aqueous solution at a flow rate of 1 mL/min; the column temperature was 30 ℃; the detection wavelength was 360 nm. The content of hyperoside, an active ingredient in SSHFG, was determined by a CAPCELL PAK C18 MGⅡ(4.6 mm × 250 mm, 5.0 μm) column with isocratic elution of acetonitrile-0.2% acetic acid aqueous solution (16∶84) at a flow rate of 1 mL/min; the column temperature was 30 ℃; the injection volume was 5 μL; the detection wavelength was 360 nm.ResultsThe characteristic fingerprint and content determination methods of SSHFG were developed based on high-performance liquid chromatography(HPLC). Six characteristic peaks, including neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, rutin, hyperoside and isoquercitrin, were identified in the characteristic fingerprint chromatogram of SSHFG. The similarity of HPLC characteris fingerprints among 10 batches of SSHFG samples was between 0.997 and 1.000, while the similarity between each batch and the control chromatogram was between 0.998 and 1.000. The content determination method of SSHFG was established using hyperoside as index ingredient. The content of hyperoside in 10 batches ranged from 0.484 mg/g to 0.549 mg/g, with an average content of 0.514 mg/g.ConclusionThe characteristic fingerprint and content determination methods established in present study are simple, accurate and reproducible, which can effectively evaluate the quality of SSHFG, and provide a reference to ensure the effectiveness of SSHFG in the clinical application.

摘要:ObjectiveTo explore the mechanism of modified Pentongling Formula on analgesic in endometriosis (EMs) model rats based on phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway.MethodsThe rat model of EMs was established by autologous endometrial transplantation. The EMs rats with pain behavior were chosen by the “Up & Down” method and randomly divided into model group, modified Pentongling low, medium and high dose group (n=8); and the sham group was set with 8 rats. The sham group and model group were given 0.9% NaCl solution enema, and each modified Pentongling group was given different doses of modified Pentongling Formula enema. After continuous enema for 4 weeks, the changes of 50% paw mechanical withdrawal threshold (50% PMWT) in rats were observed. HE staining was used to observe the pathological changes of the endometrium in rats, immunohistochemistry was used to detect the protein expression levels of PI3K and Akt in the ectopic lesion, ELISA was used to detect the concentrations of estradiol (E2) in the peritoneal fluid and brain-derived neurotrophic factor (BDNF) in the serum, and RT-qPCR was used to detect the mRNA expression levels of PI3K and Akt in the ectopic lesion.Results①After modeling, the 50% PMWT of rats in each modeling group significantly decreased (P<0.05). Compared with the model group, the 50% PMWT of the modified Pentongling groups significantly increased after 4 weeks administration (P<0.001). ②HE staining showed that the epithelial cells were flat, the glands significantly decreased or even disappeared, blood vessels were abundant, and there was inflammatory cell infiltration in the stroma in the endometrial tissue of the model group. Each dose of modified Pentongling could improve the pathological morphology of endometrial tissue to different degrees, and the high dose had the best effect. ③Compared with the sham group, the expression of E2, BDNF was up-regulated in the model group (P<0.01, P<0.001). Compared with the model group, E2 and BDNF expression was down-regulated in modified Pentongling each group (P<0.05, P<0.001). ④Compared with the sham group, PI3K and Akt mRNA and protein expressions in the model group was up-regulated (P<0.05, P<0.01). Compared with the model group, the Akt protein expression was down-regulated in modified Pentongling each dose group (P<0.05, P<0.01), the Akt mRNA expression was down-regulated in modified Pentongling high dose group (P<0.05), and the PI3K mRNA and protein expressions were down-regulated in modified Pentongling medium and high dose groups (P<0.05, P<0.01). ⑤Correlation analysis showed that the expression of E2, BDNF and the mRNA and protein expression of PI3K and Akt were negatively correlated with 50% PMWT.ConclusionModified Pentongling Formula may relieve EMs pain by inhibiting E2 to regulate PI3K/Akt signaling pathway.
摘要:ObjectiveTo explore the mechanism of phenylethanoid glycosides against Alzheimer’s disease (AD) based on amyloid β-protein (Aβ).MethodsTaking the natural product library as the screening object, the Autodock software was used to screen out compounds that were well docked with β-secretase 1 (BACE1) from the library by docking method. Grid Score was used as the selection standard to select candidate compounds. The affinity of these compounds with BACE1 at the concentration of 10 μmol/L was tested, and their structural characteristics were analyzed, and 1~2 compounds were finally selected as target molecules. Western blot was conducted to detect the expression levels of Aβ related proteins.ResultsEight natural products were screened out according to Grid score from high to low. Among them, the binding affinity of 3 compounds with BACE1 was more than 60% at 10 μmol/L. The structural analysis showed that 2 compounds belonged to phenylethanoid glycoside. Plantamajoside and forsythiaside were finally selected as the target compounds. The IC50 of plantamajoside and forsythiaside against BACE1 were (3.86±0.21) μmol/L and (4.52±0.29) μmol/L, respectively. The expression levels of BACE1 and sAPPβ in rat neurons were significantly down-regulated by plantamajoside and forsythiaside(P<0.05, P<0.01, P<0.001), while the expression of α-secretase (ADAM10) was significantly up-regulated (P<0.05, P<0.01).ConclusionThe phenylethanoid glycosides plantamajoside and forsythiaside can inhibit the expression of Aβ by down-regulating the expressions of BACE1 and sAPPβ and up-regulating the expression of ADAM10, which slows down the development of AD. And this provides the high quality of lead compounds for the research of Aβ pathway against AD.
摘要:Objective: To observe the therapeutic effects of different formulas of Sihuang Powder Ointment topically applied on mice with fluorouracil-associated phlebitis, and to select the optimal formula for the treatment of phlebitis.MethodsDifferent formulas of Sihuang Powder Ointment A, B, C, D were prepared. Forty-nine mice were randomly divided into 7 groups: blank group, model group, positive control group and Sihuang Powder Ointment A, B, C, D groups. Except for the blank group, the mice in other 6 groups were injected with fluorouracil through tail vein for 3 consecutive days to establish phlebitis model. The Sihuang Powder Ointment groups were treated topically with Sihuang Powder Ointment A, B, C, and D correspondingly, the positive control group was treated topically with mucopolysaccharide polysulfate cream, and the blank and model groups were treated topically with blank excipient. The degree of phlebitis, the swelling degree of rat tail, the levels of inflammatory factors in venous blood, and the rat tail pathomorphological changes were observed in each group.ResultsCompared with the blank group, the grade of phlebitis, swelling degree of tail, and levels of inflammatory factors in venous blood in the model group were increased (P<0.05, P<0.001), indicating successful molding. After external treatment with Sihuang Powder Ointment A, B, C, D for 5 d correspondingly, the grade of phlebitis and swelling degree of tail in each Sihuang Powder Ointment group were decreased compared with those before treatment (P<0.05, P<0.001), the levels of CRP, IL-6 and TNF-α were decreased compared with the model group (P<0.001). The levels of CRP and IL-6 were decreased in Sihuang Powder C group after treatment compared with the positive control group (P<0.05, P<0.001). The degree of phlebitis inflammation in each Sihuang Powder Ointment group was reduced based on morphological observation of pathological sections.ConclusionExternal application of different formulas of Sihuang Powder can effectively reduce the phlebitis in mice induced by intravenous injection of fluorouracil, among which Sihuang Powder C has the best effect.
摘要:Objective: To analyze the characteristics and internal mechanism of Xue Shengbai’s prescriptions for the differentiation and treatment of gastrointestinal diseases in Xue Xue’s Medical Cases based on the traditional Chinese medicine inheritance auxiliary platform (TCMIAP) and network pharmacology, thus providing reference for traditional Chinese medicine diagnosis and treatment of digestive system diseases represented by damp-heat gastrointestinal diseases.MethodsMedical cases related to differentiation and treatment of gastrointestinal diseases in Xue Xue’s Medical Cases were screened, and these data were standardized and entered into TCMIAP to analyze the drug frequency, prescription rules, etc. The core drug combination was obtained and analyzed by protein-protein interaction (PPI) analysis, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis, to explore its mechanism in the treatment of gastrointestinal diseases.Results①A total of 191 medical cases of differentiation and treatment of gastrointestinal diseases in Xue Xue’s Medical Cases were included, involving 219 traditional Chinese medicines. According to the drug frequency, prescription rules and new prescription analysis, the core drug combination was confirmed as follow: Magnoliae Officinalis Cortex, Tsaoko Fructus, Ginseng Radix Et Rhizoma, Poria, Citri Reticulatae Pericarpium, Arecae Pericarpium, Atractylodis Macrocephalae Rhizoma, Pogostemonis Herba. ②PPI analysis showed that proteins with high degree values included protein kinase Bα (Akt1), tumor necrosis factor (TNF), tumor protein p53 (TP53), vascular endothelial growth factor A (VEGFA), proto-oncogene tyrosine-protein kinase Src (Src), heat shock protein 90 α family class A member 1 (HSP90AA1), etc. And pathways such as phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway, hypoxia inducible factor-1 (HIF-1) signaling pathway, VEGF signaling pathway, cancer pathway were involved.ConclusionBased on the analysis and summary of Xue Xue's Medical Cases, the core drug combination for the differentiation and treatment of gastrointestinal diseases was summarized, which has the function of drying dampness by bitter herbs and resolving dampness by aromatic herbs, invigorating spleen and regulating qi, and exerts therapeutic effects such as anti-inflammation, anti-tumor, anti-oxidation, anti-bacteria through multi-component, multi-target and multi-pathway.

摘要:Through the objective analysis of the original text of Treatise on Febrile Diseases, the dialectical thoughts of Zhang Zhongjing’s symptom speculation and prediction are discussed. It includes differentiating the pulse and symptoms of recovery, predicting the concomitant symptoms of recovery, speculating the due symptoms with pulse patterns, predicting the symptoms caused by therapeutic error, prejudging and differentiating unknown pulse and symptoms based on the known ones, prejudging jaundice symptoms, mutual prejudging urine and bowel symptoms, and so on. It has important guiding value for the research and application of traditional Chinese medicine syndrome differentiation theory and the improvement of clinical efficacy.

摘要:Jujuboside A (JuA) is the main component of saponin in Ziziphi Spinosae Semen. Domestic and foreign studies have shown that JuA has a wide range of pharmacological effects, such as sedation, sleep improvement, neuroprotection, memory improvement, heart protection, anti-oxidation, anti-inflammation, and anti-tumor. With the update of pharmacological research methods and techniques, the pharmacokinetics, pharmacological effects and related molecular mechanisms of JuA have been continuously discovered and further investigated. In this paper, the research progress of pharmacokinetics and pharmacological actions of JuA was elaborated, which can provide references for future researches.
摘要:This paper reviews the mechanisms of massage in treating diseases through neural circuits. Neural circuit is the structural basis for the coordination of advanced neurological functions such as sensation, movement, balance and cognition, and is the functional unit of neural development, remodeling and rehabilitation. This paper explores the mechanisms of massage in treating diseases through neural circuits based on the influence of multi-target and provides a scientific evidence for the clinical application and research of massage.
