      摘要:Alzheimer’s disease(AD) is one of the most common neurodegenerative diseases during human aging. With the increasing aging of the world’s population, the incidence of AD is also increasing, which brings serious health threat and economic burden to patients’ families and society. In recent years, numerous studies have found that histone acetylation and deacetylation, as an epigenetic modification, play an important regulatory role in the pathogenesis of AD. Histone deacetylases (HDACs) can deacetylate histone in cell, leading to chromatin contraction, thereby inhibiting gene transcription, regulating protein homeostasis, affecting cell metabolism, aging, apoptosis and other life processes. The role and function of the HDACs family in the pathogenesis of AD and the research overview of related HDACs inhibitors in AD are summarized to provide new ideas for understanding the biological regulation mechanism mediated by the HDACs family and important theoretical guidance for the treatment of AD with HDACs inhibitors.

      摘要:Objective: To observe the clinical efficacy of Tiaogan Jieyu Decoction in the treatment of perimenopausal emotional disorder, and preliminarily explore its mechanism based on neuropeptide regulation.MethodsA total of 101 patients of perimenopausal emotional disorder with liver stagnation and blood deficiency type were selected and randomly divided into the treatment group (51 cases) and control group (50 cases). The patients in both groups were treated with basic psychological counseling. Meanwhile, the patients in the treatment group were orally treated with Tiaogan Jieyu Decoction and the patients in the control group were orally treated with Gengnianning. The treatment course of both groups was 8 weeks. Before and after treatment, the total scores of Chinese medical symptoms and main symptom scores between the two groups were compared, and the scores of Greene menopause scale, self-rating anxiety scale (SAS) and self-rating depression scale (SDS) of the two groups were evaluated and compared. Before and after treatment, the levels of estradiol (E2), follicle stimulating hormone (FSH), luteinizing hormone (LH), testosterone (T), progesterone (P), prolactin (PRL) and β-endorphins (β-EP), serotonin (5-HT), norepinephrine (NE) and dopamine (DA) were detected and compared.Results①After treatment, the total scores of Chinese medical symptoms in the two groups were significantly decreased compared with those before treatment (P<0.01), and the score in the treatment group was significantly lower than that in the control group (P<0.01). ②After treatment, the scores of main symptoms (such as tidal fever and sweatiness, depression, etc) in the two groups were decreased compared with those before treatment (P<0.05, P<0.01), and the scores of main symptoms (such as tidal fever and sweatiness, depression, anxiety, etc) in the treatment group were lower than those in the control group (P<0.05, P<0.01). ③After treatment, the scores of Greene menopause scale in the two groups were significantly decreased compared with those before treatment (P<0.01), and the score of the treatment group was significantly lower than that of the control group (P<0.01). ④After treatment, the scores of SAS and SDS in the two groups were significantly decreased compared with those before treatment (P<0.01), and the scores of the treatment group were lower than those of the control group (P<0.05). ⑤After treatment, the levels of E2 and P in the two groups were increased (P<0.05, P<0.01), the level of FSH was decreased (P<0.01), the level of LH in the treatment group was also decreased (P<0.05), and the levels of E2 and P in the treatment group were higher than those in the control group (P<0.05), the level of FSH was lower than that in the control group (P<0.05). ⑥After treatment, the level of β-EP in the two groups was decreased compared with that before treatment (P<0.05, P<0.01), the level of 5-HT was increased (P<0.05), and the level of β-EP in the treatment group was lower than that in the control group (P<0.05).ConclusionTiaogan Jieyu Decoction can effectively improve the clinical symptoms, anxiety and depression status in patients of perimenopausal emotional disorder with liver depression and blood deficiency type, and its mechanisms may be related to the regulation of neuropeptide levels such as neuroendocrine hormones, β-EP and 5-HT.

      摘要:Objective: To observe the clinical efficacy of Xiaoliu Forluma in the treatment of diffuse large B-cell lymphoma (DLBCL) and its influence on the expressions of cytokines, miR-532-3p and β-catenin mRNA.MethodsSixty patients of DLBCL were included and randomly divided into the control group and treatment group, 30 cases in each group. Meanwhile, 15 healthy volunteers were included as the normal group. The patients in the control group were treated with routine R-CHOP chemotherapy, and the patients in the treatment group were treated with modified Xiaoliu Forluma orally based on the treatment of the control group. The treatment course of both groups was 4 chemotherapy cycles. The clinical efficacy and the efficacy of traditional Chinese medicine syndrome in both groups were evaluated. The levels of serum interleukin (IL)-2, IL-4, IL-6 and IL-10 and the expression levels of miR-532-3p and β-catenin mRNA in peripheral blood lymphocyte of all the subjects were detected.Results①After treatment, the total clinical effective rate was 93.3% in the treatment group and 80.0% in the control group, and the effect of the treatment group was superior to that in the control group (P<0.05). ②After treatment, the total effective rate on traditional Chinese medicine syndrome was 96.7% in the treatment group and 83.3% in the control group, and the traditional Chinese medicine syndrome effect of the treatment group was superior to those in the control group (P<0.01). ③Compared with the normal group, the expression level of miR-532-3p in the two groups before treatment was significantly decreased (P<0.01), and the expression level of β-catenin mRNA was significantly increased (P<0.01). After treatment, the expression level of miR-532-3p in the two groups was increased (P<0.01), the expression level of β-catenin mRNA was decreased (P<0.01), and the expression level of miR-532-3p in the treatment group was higher than that in the control group (P<0.01), the expression level of β-catenin mRNA was lower than that in the control group (P<0.01). ④Compared with the normal group, the levels of IL-4, IL-6 and IL-10 in the two groups before treatment were significantly increased (P<0.01), and the level of IL-2 was significantly decreased (P<0.01). After treatment, the levels of IL-4, IL-6 and IL-10 in the two groups were significantly decreased (P<0.01), the level of IL-2 was significantly increased (P<0.01), and the levels of IL-4, IL-6 and IL-10 in the treatment group were lower than those in the control group (P<0.05), the level of IL-2 was higher than that in the control group (P<0.05).ConclusionXiaoliu Formula combined with chemotherapy can effectively inhibit the tumor and improve the clinical symptoms and the tumor microenvironment, and its mechanisms may be related to regulating the expression of miR-532-3p and β-catenin mRNA.

      摘要:Objective: To explore the influencing factors of Huachansu Injection intraperitoneal perfusion in treating patients of colon cancer with malignant ascites and summarize the characteristics of the benefit population, in order to better guide the clinical medication.MethodsA retrospective study was applied, and the clinical data of 135 patients of colon cancer with malignant ascites treated by Huachansu Injection intraperitoneal perfusion were collected. Taking the ascites volume control efficacy and the ascites color improvement efficacy as the outcome indexes, the logistic regression analysis was conducted to investigate the influences of the factors including gender, age, ascites property, Chinese medicial syndrome and other factors on the efficacy.ResultsAmong the 135 patients, the ascites volume control was effective in 83 cases and the ascites color improvement was effective in 94 cases. The results of logistic regression analysis showed that, the primary location of colon cancer, ascites property and combining with systemic Chinese medicine treatment were the independent influencing factors on the ascites volume control efficacy, and liver metastasis, ascites property, increased bilirubin and local syndrome differentiation were the independent influencing factors on the ascites color improvement efficacy.ConclusionHuachansu Injection intraperitoneal perfusion has better effect in the treatment of colon cancer with malignant ascites for the patients whose primary location are left colon, without liver metastasis and increased bilirubin, with bloody ascites and dampness-heat toxicity syndrome of local syndrome differentiation, treated by combining with systematic Chinese medicine. Through the preliminary screening of population characteristics before treatment, the clinical efficacy might be improved.

      摘要:Objective: To observe the clinical effect of Tiaozhong Baoyuan Decoction in the treatment of delayed healing of long bone fracture after operation.MethodsA total of 60 patients with delayed healing of long bone fracture after operation were included and randomly divided into the treatment group and control group, 30 cases in each group. The patients in the control group were treated with Alfacalcidol Soft Capsule orally and the patients in the treatment group were treated with Tiaozhong Baoyuan Decoction orally. The treatment course of both groups was 6 months. Before treatment, three months and six months after treatment, the fracture connection of the patients in the two groups was evaluated by fracture healing imaging score (RUST score). Three months and six months after treatment, the clinical effects of the two groups were evaluated and compared. Before treatment and 6 months after treatment, the health status of the patients in the two groups was evaluated by the MOS item short from health survey (SF-36) scale.Results①Three months and six months after treatment, the RUST scores of the two groups were increased compared with those before treatment (P<0.05), and the score of the treatment group was higher than those of the control group (P<0.05). ②Three months and six months after treatment, the total clinical effective rates of the treatment group were 73.33% and 86.67% respectively, the total clinical effective rates of the control group were 40.00% and 53.33% respectively, and the clinical efficacy of the treatment group was better than that of the control group (P<0.05). ③Six months after treatment, the scores of physical function, role physical, social function, bodily pain and general health dimension score in the treatment group were significantly increased compared with those before treatment (P<0.05), the scores of physical function, role physical and bodily pain dimension score in the control group were also significantly increased compared with those before treatment (P<0.05), and the scores of physical function, role physical, social function, bodily pain and general health dimension score in the treatment group were significantly higher than those in the control group (P<0.05).ConclusionTiaozhong Baoyuan Decoction can promote the callus growth and fracture healing, and effectively improve the health status of patients with delayed healing of long bone fracture after operation.

      摘要:Objective: To explore the mechanism of antigen-presenting B cells (APBCs) proliferationand activation.MethodsIn this study, flow cytometric sorting was used to filter the CD20+CD80+CD86+ positive APBCs. lncRNA RP11 knock-down (KD) and over-expressed (OE) cells were constructed, and combined with in vivo experiments, the regulation and molecule mechanism of lncRNA RP11 on APBCs activation was investigated.ResultsThe expression of lncRNA RP11 was increased significantly after APBCs activation. Low expression of lncRNA RP11 could significantly inhibit the activation of APBCs and secretion of IgG and IgM by B cells. While the number of activated APBCs was significantly increased and secretion of IgG and IgM was promoted in over-expressed lncRNA RP11 cells. At the same time, PI3K/Akt/mTOR pathway was activated in over-expressed lncRNA RP11 cells, but the pathway was significantly inhibited in knock-down lncRNA RP11 cells. The results of mTOR inhibitors tests confirmed that the activity level of APBCs was down-regulated because of the inhibition of the PI3K/Akt/mTOR pathway. On the basis of neuromyelitis optica spectrum disease (NMOSD) modeling, mTOR activator (MHY485) was used to activate mTOR signaling pathway, the mice inflammatory response increased and the activation degree of APBCs significantly promoted.ConclusionLncRNA RP11 can influence B cell activity by regulating PI3K/Akt/mTOR pathway.

      摘要:Objective: To explore the mechanism of Yiqi Formula enhancing the inhibiting function of cisplatin on triple-negative breast cancer.Methods①The expression levels of lncRNA HCP5 in breast cancer tumor tissues, triple-negative breast cancer tumor and tumor adjacent tissues were compared using TCGA database. ②The expression levels of lncRNA HCP5, PTEN and p-Akt protein in triple-negative breast cancer tumor and tumor adjacent tissues and non-triple-negative breast cancer tumor tissues were detected. ③The xenograft model of nude mice with triple-negative breast cancer was established and treated with cisplatin, Yiqi Formula + cisplatin. Tumor volume, weight and expression levels of lncRNA HCP5, PTEN and P-Akt protein in the tumor were measured.Results①The data of TCGA showed that the expression level of lncRNA HCP5 in breast cancer tumor tissues was higher than that in tumor adjacent tissues, but the difference was not statistically significant (P>0.05). In Basal-like breast cancer, the expression level of lncRNA HCP5 in tumor tissues was significantly higher than that in tumor adjacent tissues (P<0.01). ②In triple⁃negative breast cancer tumor tissues, the expression level of lncRNA HCP5 was higher than that in tumor adjacent tissues and non-triple-negative breast cancer tumor tissues (P<0.01), the expression level of PTEN protein was lower than that in tumor adjacent tissues and non-triple-negative breast cancer tumor tissues (P<0.001), and the expression level of p-Akt protein was higher than that in tumor adjacent tissues and non-triple-negative breast cancer tumor tissues (P<0.001).③Compared with cisplatin group, the tumor volume and weight of Yiqi Formula + cisplatin group was significantly smaller and lighter (P<0.001), the expression level of lncRNA HCP5 and p-Akt protein decreased(P<0.001), the expression level of PTEN protein increased(P<0.001).ConclusionYiqi Formula may increase PTEN protein expression and decrease p-Akt protein expression by down regulating lncRNA HCP5, so as to enhance inhibiting effect of cisplatin on triple-negative breast cancer.

      摘要:Objective: To explore the improvement effect and potential mechanism of celastrol (CEL) on glycolipid metabolism disorders in model mice by 1H-NMR metabolomics method.MethodsThirty-two male healthy C57BL/6J mice were randomly divided into control group (LFD group), model group (HFD group), control treatment group (LFD+CEL group), and model treatment group (HFD + CEL group). Mouse models of glycolipid metabolism disorders were induced by a high-fat diet. After 10 weeks, the mice in LFD + CEL group and HFD + CEL group were injected i.p. with CEL (100 μg/kg); while those in LFD group and HFD group were injected i.p. with equal amount of solvent, the administration lasted continuously for 6 weeks. The biochemical parameters were detected and histopathological changes of the mice in each group were observed. 1H-NMR metabolomics method was applied to analyze the plasma metabolic characteristics and the intervention effect of CEL in model mice.ResultsThe levels of biochemical parameters of the mice plasma in the HFD+CEL group were significantly different from those in the HFD group (P<0.01, P<0.05). Hepatic steatosis was significantly improved. Glycolipid metabolism disorders and plasma metabolism characteristics were significantly improved. The improvement effect of CEL on glycolipid metabolism disorders in model mice may be related to multiple metabolic pathways, such as lipid metabolism, creatine metabolism, β-oxidation of fatty acids, tricarboxylic acid cycle, amino acid metabolism, gluconeogenesis and glycolysis.ConclusionCEL may improve high-fat-induced glycolipid metabolism disorders by regulating lipid metabolism, creatine metabolism, β-oxidation of fatty acids, tricarboxylic acid cycle, amino acid metabolism, gluconeogenesis and glycolysis.

      摘要:Objective: To explore the common material basis of volatile oil from Acori Tatarinowii Rhizoma and mechanism in intervening Alzheimer’s disease, Parkinson’s disease, epilepsy and depression based on network pharmacology.MethodsThe volatile oil components of Acori Tatarinowii Rhizoma were obtained from TCMSP, PubChem and CNKI databases. And the components with high absorption, blood-brain barrier and drug-like properties were screened by SwissADME platform. The active components and potential targets in the treatment of 4 diseases were predicted through SwissTargetPrediction, BATMAN-TCM, GeneCards and OMIM databases. Thus, the “components-targets-diseases” network was constructed by Cytoscape 3.6.0. The protein-protein interaction (PPI) network of potential targets was constructed based on the STRING and BioGRID databases. The GO function annotation and KEGG enrichment analysis of potential targets were performed by Bioconductor database and R 4.1.2 software. Finally, AutoDock Vina was used to validate the molecular docking of key targets and key components.ResultsTotally 33 volatile oil active components from Acori Tatarinowii Rhizoma and 61 corresponding targets were obtained. The “components-targets-diseases” network showed that α-cyperone, acorenone, aristolone, isoshyobunone, acoronene, shyobunone, zierone and methyl isoeugenol were the key components in disease intervention. PPI network showed that AKT1, TP53 and mTOR were the key targets. The molecular docking results showed that the binding energies of key targets and key compounds were less than -4.7 kcal/mol. KEGG enrichment results showed that dopaminergic synapse pathway, Parkinson’s disease pathway, serotonergic synapse pathway, Alzheimer’s disease pathway were the key pathways.ConclusionEight volatile oil active components were screened from Acori Tatarinowii Rhizoma, which may exert the therapeutic effects on central nervous system diseases by participating in multiple pathways through regulatory targets AKT1, TP53 and mTOR.

      摘要:objective: To establish a UPLC assay method for the determination of gallic acid and glucose times nucleoside and a UPLC fingerprint analysis method for total phenol of Phyllanthi Fructus.MethodsThe analysis conditions were set at a CAPCELL PAK C18 MG(4.6 mm × 150 mm,5 μm)chromatographic column; mobile phase: methanol-2% acetic acid aqueous solution, gradient elution; detection wavelength: 273 nm; column temperature: 30 ℃; flow rate: 0.8 ml/min.ResultsGood linear relationship was observed for gallic acid in the range of 5.82~465.20 μg/ml and glucose times nucleoside in the range of 12.38~793.00 μg/ml respectively. And the average recovery rates of gallic acid and glucoside were 100.57% and 99.22% respectively, with the RSDs both less than 3.0%. The fingerprint showed 7 characteristic peaks, and the similarity of fingerprint was more than 0.90.ConclusionThis method is simple, accurate and reproducible, and could be used as a quality control method for total phenol of Phyllanthi Fructus.

      摘要:Objective: To establish a determination method for berberine hydrochloride and its in vitro release of berberine hydrochloride thermosensitive hydrogel based on nano graphene oxide (BH-NGO-gel).MethodsThe high performance liquid chromatography method was used, performed on a C18 column (4.6 mm × 250 mm, 5 μm) at a flow rate of 1 ml/min with mobile phase of acetonitrile-0.03 mol/L monobasic potassium phosphate (28∶72, v/v). The detection wavelength was 350 nm, the column temperature was 30 ℃ and the injection volume was 10 μl. The release medium was 0.1 mol/L phosphate buffer (PBS, pH 7.4) containing 0.5% Tween 80. The in vitro release tests were carried out by dialysis method at 37 ℃ and 42 ℃ respectively. And the kinetic model of drug release was analyzed by DDSolver software.ResultsThe linear range of berberine hydrochloride was 2~20 μg/ml, and the recoveries of high, medium and low concentrations were all more than 95%, and RSD were all less than 2%. Compared with berberine hydrochloride solution, the drug release rate of BH-NGO-gel was significantly slower (P<0.001), and the drug release rate at 42 ℃ was higher than that of 37 ℃, but there was no significant statistical difference (P>0.05). The in vitro release processes of BH-NGO-gel at 42 ℃ and 37 ℃ were all accorded with Makoid-Banakar model, and the correlation coefficients r was 0.993 6 and 0.986 9, respectively.ConclusionThe established method for determining berberine hydrochloride in vitro release from BH-NGO-gel is simple and feasible. BH-NGO-gel had obvious sustained release effect and temperature dependent drug release characteristics.

      摘要:Objective: To investigate the in vitro and in vivo recovery rates and stability of protocatechuic aldehyde brain microdialysis probe.MethodsThe concentration of protocatechuic aldehyde in microdialysis samples was determined by high performance liquid chromatography. The effects of different drug perfusion speeds and concentrations on forward and reverse recovery of protocatechuic aldehyde microdialysis brain probe in vitro and in vivo, and the stability of recovery in vitro and in vivo were investigated.ResultsThe in vitro recovery of the protocatechuic aldehyde brain probe was basically the same, and the relative recoveries of the in vitro and in vivo dialysis methods were also basically the same, so the in vivo reverse recovery can be used as the probe recovery of the drug for calibration. The recovery rate of the probe was inversely proportional to the flow rates (1.0, 1.5, 2.0, 2.5, and 3.0 μl/min) and has nothing to do with the concentration of protocatechuic aldehyde (3.13, 6.25, 12.50, 25.00 μg/ml). The in vivo and in vitro recovery rate of protocatechuic aldehyde brain probes within 8 h remained basically stable.ConclusionsThe established protocatechuic aldehyde brain microdialysis method can be used for brain pharmacokinetic study in rats.

      摘要:Objective: To study the effect of Shi’s Fumigation and Bath Formula (SFBF) medicinal bath on the hemodynamics of femoral artery in rabbits with knee osteoarthritis (KOA) and explore the appropriate treatment temperature.Methods①Sixty New Zealand rabbits were randomly divided into high-temperature group, medium-temperature group, low-temperature group, sham-operated group and model group. The KOA model was established by injecting papain and levocysteine into the right knee respectively. Four weeks after modeling, the knees of rabbits in each group were treated with pure water bath temperature gradient intervention for a total of 4 weeks to explore the appropriate treatment temperature. ②Fifty rabbits were randomly divided into SFBF medicinal bath group, pure water bath group, votalin treatment group and model group to further study the effect of traditional Chinese medicine medicinal bath on the hemodynamics of femoral artery in KOA rabbits at different temperatures based on above optimal water bath intervention temperature.Results①The intervention effect of pure water bath at 40 ℃ was optimal (P<0.05) on hemodynamic indexes such as femoral artery internal diameter, systolic peak flow rate, diastolic peak flow rate and vascular resistance index. ②Compared with medicinal bath at other temperatures and votalin treatment, the improvement of hemodynamic indexes on KOA rabbits of the SFBF medicinal bath at 40 ℃ was most significant (P<0.05).ConclusionSFBF medicinal bath can accelerate the blood flow rate of the femoral artery in KOA rabbits, reduce vascular resistance and improve local blood circulation. And 40 ℃ is the optimal temperature for the treatment of KOA with the SFBF medicinal bath.

      摘要:Based on ancient Chinese literature, the etiology and pathogenesis of dysgeusia was analyzed and combined with the characteristics of dysgeusia caused by COVI-19, its etiology and pathogenesis were concluded to be closely related to the spleen, stomach and membrane source, and dampness turbidity stasis was the key factor of pathogenesis. Exploring the treatment methods and prescriptions for dysgeusia in ancient literature, the treatment theory of dysgeusia of COVID-19 was based on eliminating dampness and turbidity. According to different syndromes, the five prescriptions of Baizhu Houpu Decoction, Ganlu Drink with patchouli, Linzhu Drink, Qingtou Emo Formula and Koudan Formula can be used respectively, or in combination with the main prescription for COVID-19, and acupuncture can be used for treatment as well.

      摘要:This study made a systematical herbal textual research on the use, source, place of origin and efficacy of ancient and modern Ophiopogonis Radix by referring to the literature of ancient herbs, and provided a basis for the selection of modern Ophiopogonis Radix varieties by combining with modern pharmacological research of traditional Chinese medicine. Through the textual research, Ophiopogonis Radix used in ancient times had a variety of sources, which is consistent with classification of modern botany. However, Ophiopogonis Radix from different sources is basically the same in use, efficacy and clinical application. According to the research result, it is not necessary to distinguish between Ophiopogonis Radix and Liriopes Radix recorded in the Pharmacopoeia of the People’s Republic of China (2020 Edition), thus these two can be merged as one so as to expand medicinal resources of Ophiopogonis Radix.

      摘要:Parkinson’s disease (PD) is a kind of neurodegenerative disease, which cannot be cured at present. In recent years, some foreign studies have applied spinal cord stimulation (SCS) to treat the disease, which is consistent with the idea of Du meridian acupuncture treatment of traditional Chinese medicine. Through systematically reviewing the main studies of SCS in the treatment of PD, its mechanisms were analyzed. Meanwhile, through analyzing the relationship among Du meridian, yang qi, brain and PD, this paper discusses the theoretical basis of treating PD from Du meridian. Then, from the perspective of SCS, this paper analyzes the treatment of PD from Du meridian of traditional Chinese medicine, in order to obtain a better therapy method in clinic.

      摘要:Hepatoma is a refractory malignant tumor. The rapid proliferation of cancer cells and the complex molecular stress network against drug intervention are the important reasons why it is refractory. It has a certain application prospect to develop comprehensive treatment methods including traditional Chinese medicine, develop new drugs (such as herbal extracts or active ingredients) and carry out research on combined drugs. This paper reviewed the researches on inhibition of proliferation of hepatoma cells by the combination of traditional Chinese medicine and its mechanism, hoping to provide reference for the comprehensive treatment of hepatoma and the related research of traditional Chinese medicine.
