目的

探讨5种柑橘属药材(青皮、广陈皮、枳实、枳壳和橘红)中辛弗林含量的差异,以期为其质量评估提供科学依据。
方法

采用Waters 超高效合相色谱技术(UPCC),Waters Trefoil™ CEL2色谱柱(3.0 mm×150 mm,2.5 μm),流动相A为CO
2
,流动相B为0.5%三氟乙酸/0.5%三乙胺-甲醇溶液,检测波长设定为224 nm,柱温为40 ℃,对5种柑橘属药材中的手性辛弗林进行检测分析。
结果

辛弗林在37.5~1 200 mg/L的浓度范围内呈现出良好的线性关系(
r
=0.999 3);其平均回收率为102.68%,相对标准偏差(
RSD
)为2.84%。在5种柑橘属药材中,
R
‑辛弗林含量的排序为青皮
>
枳实
>
广陈皮
>
橘红
>
枳壳。
结论

该研究首次采用超临界流体色谱技术分离柑橘属药材中的手性辛弗
林,对其中的
R
-辛弗林进行含量测定,并且使用分子对接技术探讨了
R
-辛弗林和
S
-辛弗林的活性差异,以期为柑橘属药材的质量评价提供帮助。
Objective: To explore the variations in synephrine content across five citrus herbs (Citri Reticulatae Pericarpium Viride, Citri Reticulatae Pericarpium, Aurantii Fructus Immaturus, Aurantii Fructus, and Citri Exocarpium Rubrum), with the goal of providing a basis for their quality assessment.
Methods

We used the Waters Ultra Performance Convergence Chromatography (UPCC). The analysis was conducted using a Waters Trefoil™ CEL2 column (3.0 mm × 150 mm, 2.5 μm). The mobile phase consisted of CO₂ as mobile phase A and a 0.5% trifluoroacetic acid/0.5% triethylamine in methanol solution as mobile phase B. The detection wavelength was set at 224 nm, and the column temperature was maintained at 40 ℃. We conducted the detection and analysis of chiral synephrine in five citrus herbs.
Results

The linearity of synephrine was optimal in the concentration range of 37.5~1 200 mg/L (
r
=0.999 3), with an average recovery of 102.68% and a relative standard deviation (
RSD
) of 2.84%. The ranking of synephrine content among the five citrus herbs is as follows: Citri Reticulatae Pericarpium Viride

This study is the first to use supercritical fluid chromatography technology to separate chiral synephrine in citrus medicinal materials, quantify the

-synephrine, and employ molecular docking techniques to investigate the activity differences between
R
‑synephrine and
S
-synephrine, with the aim of providing insights for the quality evaluation of citrus herbs.
